Why do I have to update the view?
After selection of the components, the spectral diagram and the form are not automatically updated. This allows you to select all the required components first and then to update the complete view. The question mark buttons behind the selection menus for dyes and filter sets will be linked to the respective info pages by the update.

What are the question marks used for in the interactive database?
Clicking on a question mark button enables you to open an additional information window as soon as you have selected a dye or a filter set and updated the view. The spectral lines of the selected dye or filter set are displayed in the information window. Furthermore, you can then select the required filter set or dye from the list of recommendations from ZEISS and have the resulting efficiency curves displayed. The recommended filter sets are assorted in such a way that the filter set with the optimum overall efficiency appears on top of the list.

What is the meaning of the different colors of the spectral lines?
The different colors are only used to allow distinction between the selected components.

What is the meaning of the checkboxes in front of the various components?
The checkboxes in front of the components enable you to influence the display in the spectral diagram. Only the curves of activated components are displayed. In most display programs, the checkbox is ticked in this case.
Important: calculation of the overall efficiency is only influenced by the selection of components, not by their being activated or deactivated.

What is total efficiency?
Total efficiency curves are used to visualize the spectral excitation and emission courses of the selected dye created by interaction between the light source, filters, an objective and a detector. The area below the total efficiency curve corresponds to the light energy for excitation and emission.

How are total efficiency curves calculated?
Total efficiency curves are calculated as a simple product from the characteristic spectral lines of the selected components. For example, if a filter set and a dye have been selected, the total efficiency curves will display the multiplication of these two spectral lines. Therefore, each total efficiency curve is smaller than each single spectral line.
Important: as the quantum efficiency – a measure for the transition from excitation to emission of a fluorescence dye – is generally not exactly known, a connection between excitation and emission has not been taken into consideration in the display. Each total efficiency curve must be viewed separately.
In the view of the interactive data base, only dye 1 and filter set 1 are considered for the calculation of the total efficiency curves. Filter set 2 and dyes 2 and 3 are not taken into consideration. You can use them for the direct comparison of components.

With some filter sets, several filters are listed in a selection menu. Why?
Some filter sets consist of more than 3 filters. For example, a filter set can contain several excitation filters. With the selection menu, you can choose the suitable filter.

Why is a filter set recommended although its total efficiency curve features only low values?
The total efficiency curves are calculated from the selected filters. However, a filter set can have several excitation filters, for example. A filter set is recommended if at least one combination of filters is suitable for the dye. Please use the selection menu to choose another filter from the selected filter set.
